To evaluate the growth curve by ATP and colony counts for a probiotic product at multiple time points on contaminated and uncontaminated stainless steel surfaces.

Experiment Procedure:
Six autoclaved stainless-steel coupons per time point were labeled as one positive (P+), two negatives (N-), test one (T1), and test two (T2). Table 1 is an overview of the coupon descriptions.

100 microliters of grape juice or TSB were applied and spread on the surface of the negatives, T1, and T2 coupons, and air-dried overnight. The P+, T1, and T2 coupons were treated with 1000μl of Culleoka product to the surface for two minutes and wiped dry with a microfiber towel. Coupons were tested at the following time points (hours): 0, 4, 8, 24, 48, 72, and 96.
ATP Surface Testing
ATP Testing was performed using the Hygiena UltraSnap Surface ATP Test Device Swabs and Hygiena SystemSure Plus Luminometer ATP Bio-Contamination Testing Meter. Swabs were removed from the fridge fifteen minutes before testing to allow them to reach closer to the room temperature. At the designated time points, a new swab was used to measure the P+, N- GJ, N- TSB, T1 GJ, and T2 TSB surfaces, and the results from the meter were recorded.
Colony Counts
Treated coupons at the designated time points were transferred with forceps into 15 ml of D/E neutralizing broth into four separate 50ml conical tubes labeled P+, N- GJ, N-TSB, T1 GJ, and T2 TSB. Conical tubes were placed on the shaker for 10 minutes. Dilution tubes were filled with 900μl of 1X PBS, and serial dilutions were made for P+, T1 GJ, and T2 TSB from 10-3 to 10-8 using 100μl of the shaken solution. The stock and serial dilution solutions were plated and spread evenly over the surface of the solid tryptic soy agar (TSA) using a metal spreader. Finished plates were placed into a clean labeled zip lock bag and incubated at 37°C overnight. Isolated colonies were counted, and colonies over 300 were determined as too numerous to count (TNTC).
Trial Results:
ATP Results:
ATP results showed that the Culleoka product did not have an ATP reading but did not completely interfere with readings when mixed with nutrients. This does not mean that there wasn't an inhibitor in the cleaner that prevented ATP readings. Based on the results below in Figure 1, ATP is not an appropriate method to determine the growth curve of the Culleoka organism.

Colony Counts:
The growth curve in Figure 2 shows that both GJ and TSB are appropriate nutritional mediums that have similar growth curves at the designated time points. There was some variance in the colony counts of GJ and TSB at the eight-hour time point. Figure 3 and Table 2 are of the recovery of probiotic bacteria when the product was used alone on a clean stainless-steel surface at the designated time points. When the product was placed on a clean surface, it kept a 6-7 Log throughout each of the time points tested.

Success Rating:
Results successful using TACT (time, agitation, concentration, and temperature, as well as rinsing and drying) and/or other cleaning chemistries examined.
Conclusion:
Culleoka did show growth over time when nutrients were introduced to the surface. The best method to verify based on this testing is using colony counts as the analytical method. ATP was not consistent with the growth seen on the plates, and there may be an inhibitor in the product slightly interfering with ATP readings.

Although you may download the data generated from your authorized use of the CleanerSolutions™, TURA Data, and P2OASys™ online tools, you agree not to download, copy, or reverse engineer any part of the CleanerSolutions™, TURA Data, and/or P2OASys™ online tools, including but not limited to, the databases of the CleanerSolutions™, TURA Data, and/or the P2OASys™ online tools.
You agree to refrain from systematic automated data collection activities (including scraping, data mining, and extraction and re-utilization) of the whole or a substantial part of the TURI website and the CleanerSolutions™, TURA Data, and/or P2OASys online tool databases.
Privacy
We may collect certain personal information from you when you submit User Content or interact with the Services, including, but not limited to, the type of device you are using, web browser, IP address, search terms, email address, phone number, institution and/or employer. We only use personal information to contact you about your User Content submission, and, may, from time to time, alert you to updates or events at TURI. You may always opt-out of further communications from us. We never sell your personal data. Because TURI is a state agency, any User Content submitted to us may be disclosed pursuant to a public records request under the Massachusetts Public Records Law, MGL c. 66.
License
By submitting User Content to TURI, you grant us a worldwide, non-exclusive, royalty-free license (with the right to sublicense) to use, copy, reproduce, process, adapt, modify, publish, transmit, display and distribute User Content in any and all media or distribution methods now known or later developed (for clarity, these rights include, for example, curating, transforming, and translating). This license authorizes us to make User Content available to the rest of the world without attribution, and to let others do the same. Content created, modified, compiled, arranged, and/or displayed by TURI and/or by TURI’s agents or representatives through the Services is copyrightable material that is exclusively owned by TURI, and is protected under and subject to all of the rights and remedies provided under the U.S. Copyright Act at 17 USC §§ 101 et seq. Therefore, unless otherwise expressly allowed for herein or unless expressly approved in writing by TURI, any copying or distribution of the Content is prohibited and is a violation of the U.S. Copyright Law and world copyright laws under, for example, the Berne Convention and the Universal Copyright Convention.
General
We may revise these Terms from time to time. The changes will not be retroactive, and the most current version of the Terms, will govern our relationship with you. By continuing to access or use the Services after those revisions become effective, you agree to be bound by the revised Terms.
The laws of the Commonwealth of Massachusetts, excluding its choice of law provisions, will govern these Terms and any dispute that arises between you and TURI. All disputes related to these Terms or the Services will be brought solely in the Suffolk County Superior Court located in Boston, Massachusetts, United States, and you consent to personal jurisdiction and waive any objection as to inconvenient forum.
If you are a federal, state, or local government entity in the United States using the Services in your official capacity and legally unable to accept the indemnification, controlling law, jurisdiction or venue clauses above, then those clauses do not apply to you.
In the event that any provision of these Terms is held to be invalid or unenforceable, then that provision will be limited or eliminated to the minimum extent necessary, and the remaining provisions of these Terms will remain in full force and effect. TURI’s failure to enforce any right or provision of these Terms will not be deemed a waiver of such right or provision.
